I think that phrase means one thing. The person who said is super insecure. That's all.

Person 2 "Dude, I love you. No homo" *laughs*
Person 1 "Dude I know what you meant, you didn't need to say no homo."
Person 2 "I just want everyone to know that I'm not a filthy faggot."
Person 1 "Jesus, just chill."
Person 2 "DUDE YOU ARE SOOO GAY! GET AWAY FROM ME!" *leaves without his homework*
